چکیده

Tethered particle motion is an experimental technique to monitor conformational changes in single molecules of DNA in real time, by observing the position fluctuations of a micrometersize particle attached to the DNA. This article reviews some recent work on theoretical problems inherent in the interpretation of TPM experiments, both in equilibrium and dynamical aspects.

Colloidal glass transition observed in confinement.

We study a colloidal suspension confined between two quasiparallel walls as a model system for glass transitions in confined geometries. The suspension is a mixture of two particle sizes to prevent wall-induced crystallization. We use confocal microscopy to directly observe the motion of colloidal particles. This motion is slower in confinement, thus producing glassy behavior in a sample which ...

Probing DNA conformational changes with high temporal resolution by tethered particle motion.

The tethered particle motion (TPM) technique informs about conformational changes of DNA molecules, e.g. upon looping or interaction with proteins, by tracking the Brownian motion of a particle probe tethered to a surface by a single DNA molecule and detecting changes of its amplitude of movement. We discuss in this context the time resolution of TPM, which strongly depends on the particle-DNA ...

Tethered particle motion as a diagnostic of DNA tether length.

The tethered particle motion (TPM) technique involves an analysis of the Brownian motion of a bead tethered to a slide by a single DNA molecule. We describe an improved experimental protocol with which to form the tethers, an algorithm for analyzing bead motion visualized using differential interference contrast microscopy, and a physical model with which we have successfully simulated such DNA...

Preparation and Characterization of Colloidal Silica in Alkaline and Constant Range of pH

In this study, alkaline colloidal silica was prepared by titration of an alkaline solution such as sodium silicate with silicic acid in constant range of pH  .This constant range of pH was achieved by using concentrated soduim silicate solutions as pH adjusting solutions during titration. The sodium silicate solutions  having a variety of...
